Quantitative Determination of Metoclopramide in Meat by Ultra-High Performance Liquid Chromatography

Abstract:An analytical method for the quantitative determination of metoclopramide in meat by ultra-high performance liquid chromatography (UPLC) was established in this paper. Samples were extracted with acetonitrile and then the extract was purified using an HLB cartridge. The chromatographic separation was accomplished on a C18 column (2.1 mm × 100 mm, 1.7 μm) using a mobile phase consisting of acetonitrile and 0.1% formic acid in water (20:80, V/V) at a flow rate of 0.3 mL/min. The detection wavelength was 275 nm and the column temperature was 35 ℃. The injection volume was 3.0 μL. The results showed that the linear range of metoclopramide was from 1 to 100 mg/L (R2 = 0.999 5). The limit of detection was 0.25 mg/kg, and the limit of quantitation was 0.5 mg/kg. The precision, expressed as relative standard derivation (RSD) was 0.79%–5.27% (n = 12). The average recovery was 87.0%–106.0%. The developed method proved to be applicable to the determination of metoclopramide in meat.
